Scheffler, who already led the FedEx Cup standings, birdied his first five holes at TPC Southwind and recorded a total of 11 birdies, with bogeys on the eighth and 12th holes.
South Korea's Im Sung-jae and Norway's Viktor Hovland are tied for second at eight under overall. Reigning FedEx Cup champion Tommy Fleetwood is the highest placed British player at six under.
Scotland's Robert MacIntyre withdrew from the event on Friday due to a knee injury. He had scored a 12-over 82 in the first round on Thursday.
